COMPARISON OF LEUKOCYTE TYPE MORPHOLOGY USING AIR CONDITIONER WASTE AND PHOSPHATE BUFFER IN PBS PREPARATION
Keywords:
Air Conditioner Waste, Giemsa Staining, Buffer FosfatAbstract
In a health laboratory, Giemsa staining is routinely performed. The Giemsa staining procedure is carried out to identify various cell types, one of which is leukocytes in PBS (a common acronym likely referring to a preparation technique, such as a smear or differential count). Before staining, the Giemsa stain must first be diluted using a buffer. The use of Air Conditioners (AC) in daily life has become common among Indonesian society. The commonly used AC often produces waste water or effluent that is directly discharged into drainage systems or dedicated pipes without being utilized for other purposes. Based on previous research, it has been shown that AC waste water has properties similar to deionized water (aquadest) and can be used as an alternative substitute for it. This study aims to determine whether there is a difference in the quality of the slides for examining leukocyte type morphology when using Air Conditioner (AC) waste as a buffer substitute in Giemsa staining. The research method is experimental, involving taking blood samples from respondents. The research data was analyzed using the Wilcoxon test. The results obtained an Asymp. sig > 0.05 for all leukocyte cell nuclei and monocyte cytoplasm. However, the cytoplasm of eosinophils, eosinophil granules, cytoplasm of neutrophils, neutrophil granules, and cytoplasm of lymphocytes received a value of < 0.05, which indicates that there is a difference in the quality of the slides for leukocyte morphology when using phosphate buffer versus AC waste The abstract should be a maximum of 250 words and contain the following substance Background: A brief explanation of the problem or issue raised in the research. Research Objective: Explain the main objective of this research. Research Method: Briefly describe the methods or approaches used in the research. Research Results: Present the main findings or results obtained from the research. Conclusion: Provide the main conclusions from the research results and their implications.
